Summary

This commentary addresses the growing understanding of the complexity in ALS causes and manifestations. For instance, there have been 120 mutations within 25 genes, identified as having a relationship with familial and non-familial ALS. Additionally, the disease can manifest itself with four different phenotypes that can progress in various ways. The next frontier in studying the causes of ALS will be research into how the factors revealed by epidemiology, genetic mutations, and cell-biology pathways might work together.
