Abstract
The mutagenic sensitivity of the arabinoside resistant (Ara-r) assay using tester (SV-50) was described for a series of 26 compounds known to be mutagenic in the Ames assay or mutagenic only in other systems. Ten of these 26 gave negative responses in the Ara-r and Ames assays both in the plate incorporation test and preincubation test. Preincubation tests enhanced the sensitivity of the Ara-r assay to 13 of 15 compounds by 2.1 to 154 times. The authors conclude that in general the mutagenic response of the Ara-r assay to the 26 compounds tested is similar to that of the Ames assay. It appeared that the preincubation test, which is a simplified combination of the suspension test and the plate incorporation test, might be a better test protocol for the Ara-r assay system with (SV- 50). The study also indicated that the Ara-r forward mutation assay using strain (SV-50) was useful for screening mutagenicity of compounds with different modes of action. The authors conclude that the Ara-r forward mutation assay using (SV-50) is a useful assay system for screening mutagenicities of compounds with different modes of action.
